Purpose and scope
Thromboxane A2 is produced from arachidonic acid by many different types of cell. It causes irreversible platelet aggregation, and vascular and bronchial smooth muscle contraction. TXA2 is rapidly hydrolysed to form thromboxane B2. The normal concentrations of TXB2 in plasma are extremely low.
11-Dehydro thromboxane B2 is derived from TXB2 by the action of 11-hydroxy thromboxane dehydrogenase. It is one of the major metabolites of TXB2 in plasma and urine. Concentrations of 11-dehydro TXB2 in plasma and urine reflect thromboxane production. In clinical trials of new medicines, 11-dehydro TXB2 serves as a pharmacodynamic measure of drug effects on platelet function.

Sample requirements
Take an aliquot of at least 10 mL from a urine collection. Immediately after collection, cool the sample in an ice bath for 20 min, and centrifuge at 4°C for 10 min at 2000G. Divide the supernatant equally between 2 polypropylene 5 mL tubes. If samples cannot be analysed immediately, freeze them at –80°C.

Purpose and scope
Prostaglandin I2 (PGI2) is formed from arachidonic acid in the vascular endothelium and renal cortex, by the activity of cyclooxygenase and PGI2 synthase. PGI2 is a potent vasodilator and inhibitor of platelet aggregation. PGI2 is non-enzymatically hydrated to 6-keto PGF1α, and then quickly converted to the major metabolite 2,3-dinor-6-keto prostaglandin F1α. Concentrations of 2,3-dinor-6-keto prostaglandin F1α in plasma and urine reflect PGI2 production. In clinical trials of new medicines, 2,3-dinor-6-keto prostaglandin F1α serves as a measure of pharmacodynamic effects on platelet function.
